Definition of active unit: when hydrolyzing BAEE at 25°C, pH 7.6, reaction system of 3.2 mL (optical path = 1 cm), one active unit will produce an increase of 0.003 in A253 (absorbance value at 253 nm) per minute.

CT26.WT and HEK293FT cells were well dissociated after 3 min of enzymolysis by Trypelectase recombinant porcine trypsin, and the cell viability results indicated minor damage to cells which will not affect cell passaging.

Usage:
Procedure of adherent cell dissociation:
Reagents and consumables: Trypsin dry powder, solving liquid, PBS (pH 7.4), 0.22 μm filter membrane
1.Take the Trypelectase trypsin dry powder reagent vial from the refrigerator and restore to room temperature (about 30 min).
2.Solving liquid: 1 mM HCl and 5% sorbitol, pH 3.0, filter through a 0.22 μm membrane for sterilization.
3.Weigh the dry powder and gently reconstitute in the solving liquid to a trypsin concentration of 10 mg/mL in the solution.
4.Preparation of the Trypelectase trypsin buffer for cell dissociation:
Mix the mother liquor with sterile PBS (pH7.4) at 1:20 (v/v) to a pancreatin cell dissociation solution of 0.5mg/mL at pH7.4. If incubation at 37°C is required, PBS can be incubated at 37°C in advance before mixing with the mother liquor.
