TrypLUS cell dissociation solution is an animal-origin free recombinant protease; as a trypsin like enzyme, it has similar dissociation kinetics to trypsin, and higher stability and purity, milder hydrolysis, and lower cytotoxicity than trypsin. It is often used in experiments involving animal cell dissociation, primary cell acquisition, or microcarrier processes.

Characterization by SDS-PAGE:
Comparison of SDS-PAGE protein gel silver staining under reducing (R) and non-reducing (NR) conditions (0.1 μg loaded) showed no heteroprotein activity data
Application case study:
TrypLUS cell dissociation solution vs. imported product: excellent dissociating performance and low cytotoxicity!
Transportation and Storage:
When stored at 2°C ~ 8°C, the validity period is 2 years.
Usage:
1.TrypLUS can be used at room temperature, or preheated to 37°C before use for better dissociating performance.
2.Procedure (using T25 flask as an example).
2.1 Start dissociation and passage when the cells cultured in T25 flask are more than 90% confluent.
2.2 Remove the cell culture medium and wash once with 10 mL PBS.
2.3 Add 1.7 mL of TrypLUS enzyme to each flask and leave to dissociate at 37°C for 3 min.
2.4 Resuspend the cells using 8.3 mL of complete medium and gently blow the cells apart using a pipette.
2.5 Measure cell count and viability with a cell counter.
2.6 Pass 1 × 106 cells into a T25 culture flask.
2.7 Observe cell adhesion 24 h after passage.
Note: Pancreatic enzyme inhibitors are not required after dissociation

Q:What is TrypLUS enzyme?
A:Chaselection TrypLUS Enzyme is a recombinant fungal trypsin like protease with functions similar to trypsin. It is an upgrade of traditional pancreatic enzyme products. Traditional pancreatic enzyme contains α- Trypsin and β- Trypsin which is not strictly uniform in terms of composition. This also leads to a decrease in the overall activity of pancreatic enzymes with the occurrence of autophagy. Therefore, pancreatic enzyme analogues with uniform components actually have a better experience in purity and specificity.
Q:What are the advantages of TrypLUS enzyme compared to trypsin?
A:1) Strong stability, stored at room temperature for a week, with minimal loss of activity.
2) High purity. it reacts with a single peak in the liquid phase, which is different from the two peaks of trypsin.
3) Mild to cells, no need to add enzyme activity inhibitors, continue passaging after use
4) Stable liquid formulations
Q: Specification and level of TrypLUS
A:We have specifications of 100ml and 500ml with two levels: GMP level and RUO level.
Q:Storage temperature and recommended storage temperature for TrypLUS enzyme?
A:In terms of performance, TrypLUS enzyme can be stored for a long time at -20 ℃, 4 ℃, and room temperature. The storage temperature of the product in the warehouse is 4 ℃.
Q:Is there a difference in the use of TrypLUS enzyme and general trypsin?
A:For customers using trypsin, TrypLUS enzyme can achieve seamless switching and consistent dissociation performance. The most important thing is that our product is ready-to-use, eliminating the need for liquid preparation and dilution
Q:What strategies can be adopted when encountering cells that are difficult to dissociate?
A:1. Extend dissociation time; 2. Digested at 37 ℃ instead of room temperature; 3. Try 10X dissociation solution
Q:What is a DMF file?
A:Drug Master Files (DMF) are information submitted by manufacturers to the US Food and Drug Administration (FDA), which includes confidential details such as product quality standards, inspection methods, production process descriptions, purity, stability, etc.
Q:What stages are DMF files applicable to customers?
A:Investigational New Drug (IND), New Drug Application (NDA), Abbreviated New Drug Application (ANDA)
Q:What is the meaning of having a DMF file?
A:Accelerate the process of drug declaration. When conducting regulatory filings for FDA clinical applications or market registration, you can directly reference our DMF number, thereby saving product review and evaluation time, reducing the preparation work for new drug clinical applications, and assisting your drug market application process.
Q:How do customers use our DMF files?
A:The DMF holder provides an authorization letter to all users who use the product, authorizing the FDA to directly review the DMF technical content involved in drug application review, without the need to provide confidential information to users
